Many people use credit cards and most of the time the credit card is not used in the right moment. I believe that credit cards are not beneficial because they aren't used for the right things. It would be very different if they were used correctly, credit cards are to be used it case of an emergency,meaning not to be used when you are going to the 7-eleven to buy a bag of chips an a soda. It has shown that more than 75% percent of americans have been bankrupt or on the verge of it.There are more than 60% of americans that have credit card debt because they are using them for the wrong things. Facts have proven that the total U. S. credit card debt, is $793. 1 Billion. and Average credit card debt per household is 15,799. Most people do not understand that when you have a credit card it comes with alot of responsibility and i say that because there are more than 10% of americans have been victims of credit card theft it may not seem like alot but credit card theft is a very serious th ing.Most complaints come from adults within the ages 40-59, Nevada, Colorado, and New Hampshire have the highest rate of credit card fraud. Having a credit card is not what people think it is; people think that if i have a credit card then i do not have to have money with me, and its just free money but its very dangerous to have a credit card. Having a credit card can lead to bankrupt and going bankrupt can make you lose everything such as your car, house, and etc. , or it could be worse an you could be placed in jail for a long time.Just because you had a credit card and used it for the wrong thing and spent way to much money†¦ A credit card is nothing but trouble each and every type of way. Most credit card companies are not looking out for your best interest. They are constantly raising interest rates. Minimum payments are just enough to cover the finance charges. Most Americans should not use credit cards for the following reasons: it will create bad spending habits; you will incur more debt affecting credit score rating; and possibly make you a victim of identity theft.In my opinion, a credit card should be used for purchases that you are able to pay off in full upon receiving your statement, but most of us don't. Most people lack self control and tend to misuse the credit card. Credit cards should mainly be used for emergencies, but we tend to use them for everyday purchases such as: food, gas, clothing, etc. Some people are living in a borrowed lifestyle, because they purchase things they can't afford. People will spend more on a purchasing using a credit card than they would with cash. The husband has a dismissive attitude towards the wife and the wife always wants more. In the story the couple has a lot of selfishness going on in their relationship. The husband ignores his wife’s needs and the wife and the wife is not satisfied with her husband. The theme of this story has a lot to do with loneliness in the couples’ relationship. In this story the American couple is trapped in their hotel room with nothing to do because of the raining weather. The husband wants to read and ignore his wife. So the wife is looking out the window and sees a cat crouched from the rain. She wants to go downstairs and get the cat. She tells her husband about it and he shrugs it off. The wife goes down herself and the story talks a lot about how she likes the inn keeper. I find that this part of the story most affected me when the husband ignored his wife’s wants and needs. She really wanted that cat because it was something for her to do and feel good about. Her husband obviously doesn’t satisfy her which makes her feel lonely and the cat would keep her company. The wife in this story wants to feel appreciated and loved like any other woman would. Being married for their first year is beginning to be hard for them. Hemingway tries to tell about a first marriage of a couple he once knew and how hard it is for some to always keep that happiness. Couples always have disagreements but should never feel alone. A husband should not ignore his wife’s needs and in this story that is what he does instead of trying to satisfy his wife. Close to the ending of this story the wife was not able to find the cat. She came back in the hotel room very sad. She tries to tell her husband but he does not listen. She starts complaining about what she wants to change about herself. She wants â€Å"her hair to be long so she can brush it† and she wants a cat. The husband can only say that he likes things the way they are. He tells her to shut up. He does not care what the wife’s needs are or what she wants. He does not want to listen and he just wants to read which is what he wishes she would do. This part of the story really interested me because I could not get as to why the husband doesn’t respond to what she asks for. The wife desperately wants attention and he is not giving it to her. The story of the American couple explains a lot about how the wife tries to get attention from the husband. Hemingway talks about a cat but behind that cat this story means more. The lack of attention her husband gives her may make her feel that she is restricted on expressing herself and keeps most of her feelings inside. The cat stays compact to stay dry outside, which is how the wife feels she has to be with her husband to make him happy. She hides herself constantly from him. In this story it relates to her as American wife and never tells her name. This kind of says that she is nothing but an American wife which is exactly how she feels. The general consensus is that the HR Department and the rest of the organization are Strange Bedfellows, when in fact they are Strategic Partners who should blend seamlessly to achieve organizational objectives. But what we see is a single-minded dislike for the HR department by most employees of the organization. The reason for this sorry state of affairs can be traced to several widespread myths regarding the function among employees. Two of the most important ones are as follows: Myth No. 1 – Employees feel that â€Å"The HR function is responsible for making employees happy. They should take care of everything, from providing employees good food to ensuring that the AC’s temperature is just right†. When in reality, an HR manager’s job is not to make employees content; it is to make them competent. In simpler words, HR Managers will not provide you a beanbag at work to recline on, but they will strive to increase your competencies that would hold you in good stead for the rest of your life. And that is all that matters in today’s professional and highly competitive world. Myth No. 2 – â€Å"The HR department comprises of a bunch of people who sit around in their ivory tower and make rules according to their whims and fancies†. While actually, HR as a function not only belongs to HR Managers, but to all managers across the organization. Translated, it means that the HR function is highly inclusive and works in tandem with all departments of the company. The criticality of the HR function can be understood by asking a simple question – What is it that makes any organization a success or a failure? Planning, Strategic decisions, Productivity, Sales figures, etc. , are a few things that instantly come to mind. What we forget to acknowledge is the fact that all these factors are a result of the efforts of the â€Å"people† of the organization. And who ensures that the right kinds of people are positioned in the right jobs in the company? The answer to that one is quite obvious. The HR department performs an array of complex activities – From Manpower Planning to Recruitment, Performance Management to Compensation and Benefits, Training and Development to Career Advancement; all of which aim at Organizational as well as individual employee development. And yet they are accused of being a ‘necessary evil’ and a ‘dark bureaucratic force’ by many. The HR function is Necessary, Yes; is it an evil? No! It is a Force, yes; but Dark and Bureaucratic? No and No! So why is HR misunderstood and mistrusted by so many, especially when the function is a benign one? Honestly, the blame lies partly with the HR managers as well for letting things get out of hand. Somewhere in the trade-off between the various roles that they are expected to play, HRs in general have started placing more importance on the roles of the ‘Admin Expert’ ‘Strategic Partner’ and have foregone the roles of the ‘Change Agent’ and ‘Employee Champion’. While they are performing the former two flawlessly, the latter roles are of great importance as well. And this negligence has resulted in widespread manifestation of disregard for the function. And unless we strike a balance between these paradoxical roles, redefining HR as an EmployeeFriendly function would be an uphill task. That said, let’s take a look what other options we have, as future HR personnel to dispel the feeling of general animosity towards the function? The answer quite simply lies in the basics. As with any other relationship, proper communication is the key to mend damages in professional relationships too. The HR person is every employee’s first point of contact before/ immediately after joining an organization. Right from the induction programs, it is the HR who facilitates the transition of a person from an outsider to a member of the organization. And we need to leverage this power for all its worth right from the word go! We need to establish proper communication channels right from the time of entry of the employee into the organization and ensure that this process of communication is a perpetual process and not an intermittent one. Next of course are the all-encompassing and highly coveted employee engagement programs. Ensuring that the HR department is projected not merely as a facilitator but also as the interface that conducts such engagement activities would ensure that employees have a change in perspective regarding HRs in general. Instead of going about this the tried and tested way, HRs can get creative and use several innovative means to achieve this. From videos depicting ‘a day in the life of an HR’ to shadowing programs (which would give employees a real-time feel of the trials and tribulations of an HR person) the possibilities are endless. And most importantly, as stressed upon earlier in this article, HR is not just about performance appraisals and pay cuts. The function is the backbone of any organization, performing an array of critical activities. HR managers need to convince the employees that they are in fact their allies and not their detractors. The new age HR managers need to remember that each action of theirs reflects on the image of the function as a whole. A slight change in our attitudes would go a long way in changing employee perceptions about our function. We as a team honestly believe that it is not a question of ‘If’ this change would happen; rather it is a question of ‘When’ it would happen. And when this happens, the HR function would assume its rightful position of importance in the Organizational structure.

They develop many characteristics and personalities such as, quantitative changes, and qualitative changes. They area key component in any childs development. There have been many scientific studies on early child development. Many theorists such as, Charles Darwin have studied and devoted a lot of time to the evolution of human development. Scientists, throughout many years have developed three separate domains. In our text, on page nine, they are listed as follows, physical development, cognitive development, and psychosocial development. Physical development is the growth of the body itself. Such as the brain, sensory capacity, and a childs motor skills. Cognitive development is the development of mental ability, learning, memory, language, thinking, moral reasoning, social and emotional growth. Psychosocial development is a pattern of changes in personality, emotions, and social relationships. Scientists categorize all of these different stages of development, but a child is constantly changing. The relationship of these developmental stages all has one common goal, and that is the development of a child into young adulthood. I prefer to use the analogy that a childs development is like a building being constructed. You start with the foundation and over time it grows into a tall, solid structure. As a parent you look at the finished product and you can be proud of the accomplishment. I don’t believe that there is one particular moment in a childs development where you can say it’s definable. It’s an ongoing process or a period of continuous development. Everything in life is a group of sequences and they are usually linked together. There are five major points of a childs development that was written in our texts on page ten. They are the prenatal period. This the period between conception and birth. Then there is the infancy to toddler hood (birth to 3yrs). Then early childhood (ages 3 to 6), middle childhood ages (6 to 11), and finally adolescence, (11 to 20 yrs). This is a very accurate process of the development and changes a child goes through. It really starts to get interesting around the middle childhood ages. This is where social construction comes in. Perceptions are created and assumptions are made by the childs peers. If the family is stable in the home, maintain a loving atmosphere and finances are stable then the child is usually well accepted. This brings me to the topic of the influences that this has on a childs development. Of course every child is an individual and has different family circumstances and environments. A childs maturation is a sequence of behavioral and physical changes. They say girls mature faster than boys. Measuring this process is a topic scientists have debated about for many years. Most believe genetics (heredity) have a lot to do with a childs maturation process. Heredity is basically inborn traits that influence the characteristics of just about every human being from conception to adulthood. Certainly the environment in which a child is brought up in has a considerable affect on a childs development. Socioeconomic status is an area we learned about in our text on pages 13 and 14; The focus was on certain factors such as, a families income, education and occupation. This part of the developmental process can be an advantage or devastating to a childs development. Many people believe if poverty is long lasting it can have a negative affect on a childs behavior and emotional state. In that aspect I agree to a certain extent. I have seen many children come from an impoverished back round and make something of themselves because there family life was strong. Parents can accept their situation or do something to make it better. If the parents are committed to the developmental process of their children it’s a matter of mental strength. Then you have children who come from an upper middle class or rich family and don’t appreciate what they have. Their parents lacked in parental skills and it showed in the way their kids developed. So I tend to think it has a lot to do with the commitment by the parents no matter what environment or socioeconomic status they are in on how their children grow up. I think it is a big mistake to assume or say it’s the norm for a child to develop properly or fail just because of their environment or parents financial status. Sigmund Freud, Erickson, Piaget, and many others have many theories on development. One thing that is perfectly clear is they a have a mutual interaction with each other. I believe the relationship is the evolution. They have many biological perspectives, theories and systems. In order to bring all of these theories together research, whether it is scientific or different perspectives must continue to be done. There are many research methods that are geared toward a childs development. We learned about two such methods and they are quantitative research, and qualitative research. One deals with measurable data and the other interprets nonnumerical data such as feelings and experiences. Research in both of these areas is widely used and provides great insight into a childs development. The difference between the two is that one is done in a controlled environment (quantitative), and the other is conducted in everyday settings, (qualitative). They both have the same common goal, which is to gather information, theorize and examine data. I personally feel like more than one mind is better. Hence the term collaborative research. This particular way of researching was talked about in our text on page fifty-one. Scientists put all their data into a pool and get different ideas and perspectives. This is a good idea and a good way to analyze and research on a broader scale. While going through this class we discussed the negative or positive affects of child care and if it was key in early childhood development. There seems to be a certain time between the ages of six and eleven where a child really starts to establish social skills. Being around other children and learning to implement their cognitive skills is essential to development. Watching how other kids react in certain situations and their behavior helps in the learning process. Piaget believes there are six stages of cognitive development and most environments a child is in all of these approaches can be implemented. Children use their brains to process information, they watch and store things in their memory for later use, and they learn from being stimulated. Processing and evaluating information is a skill they learn to hone as they continue to develop. Some say that even as an infant babies are born with an innate ability to casually reason. Infancy is a time where many emotions are being developed. Babies smile, frown, cry, laugh, and display certain temperaments. These are basic emotions, but these are the first real signs of development in a neurological sense. Infants show other signs of the way they are feeling from facial expressions motor activity and body language. In our text on pages 219 and 220 Carroll Izard wrote about these basic emotions and self conscious emotions. She and her colleagues believed facial expressions such as showing sadness, joy, fear, and interest went along way in understanding the emotional aspects of an infants development. I know as a parent that basic emotions from an infant are really a key component in understanding a babies needs and wants. Infancy is probably the time when development is a critical element. Infants need to be able to trust their caretaker. They have attachment issues and infants can easily become a little disorientated. They start to develop certain temperaments and it can have a direct impact on a parent or parents. Some of these effects can have long term results and lead to anxiety issues. Infants need to feel secure not only around his or her parents but they need the parents to maintain stability in their environment. Stability is important in the bonding process of an infants development. On page 228 and 229 of our text, stability, according to a study on 15 month old children conducted by (Jacobson Hoffman) stated that secure and less stressed toddlers were more varied and had a larger vocabulary. They also have more positive experiences with peers. On the other side of the coin the less secure toddlers showed more fear, anger, and stress. In a perfect world every parent would like the development of their children to go as smooth as possible and all of the researchers, scientists, and theorists will continue to do the best they can at giving parents the tools and knowledge to do the best they can in helping to develop their children. However, they cannot control the environment, social status, or financial status a child is born in to. Conditions change rapidly not only for children but parents as well. Sometimes there can even be internal problems that can affect the development and the relationships between all three domains of development. Yet, all or most of these potential problems can be fixed with one word and that word is stability. In this world we live in all we can ask for as parents is that our children always continue to grow and develop throughout their lifetime. Conclusion Conception all the way to adulthood is a period of great learning and development. Development is a never ending process. We are born, go through life, and then we die. It’s that simple. Life is a series of developments that never truly ends. It starts from infancy to early adolescence. Every stage is a journey, a learning experience, and a series of developments in the maturation of children. Developmental domains and developmental theories help scientists answer many questions in the development of children. As long as we keep making babies we will give theorists and scientists many more things to research and debate on, and as parents we can continue to help develop our children.

Properties of Poly(B-amino Ester)s

Properties of Poly(B-amino Ester)s The poly(b-amino ester)s, a class of biodegradable cationic polymers, were firstly  prepared by Chiellini in 198340. These polymers were based on poly(amidoamine)s  developed in 1970 by Ferruti41, that contain tertiary amines in their backbones and  can be synthesized by a simple Michael addition reaction of bifunctional amines  and bisacrylamides. However, the interest over the use of poly(b-amino ester)s rised  significantly after its use as transfection reagent at Langer Lab in 200042. The development  of poly(b-amino ester)s emerged by the need to develop a cationic polymer  for gene delivery with high transfection efficiency and long-term biocompatibility  including hydrolyzable moieties easily degradable into non-toxic small molecule  byproducts. The synthesis of this polymer can easily be accomplished: without  necessity of independent preparation of specialized monomers; the use of stoichiometric  amounts of expensive coupling reagents, or amine protecti on strategies prior  to polymerization42. The main general objective of the work of mentioned research  group was to develop a polymer-based non-viral vector more efficient and less cytotoxic  than other cationic polymers used at that time for this purpose (such as,  polyethylenimine (PEI) or poly(L-lysine) (PLL)). In fact, poly(b-amino ester) approach exhibited a particularly attractive basis for  the development of new polymer-based transfection vectors for several reasons: the  polymers contain the required amines (positive charges to complex genetic material);  readily degradable linkages (by hydrolysis of ester bonds in the polymer backbones  may increase the biodegradability and biocompatibility); and multiple analogues  could be synthesized directly from compounds commercially available (easy and inexpensive  synthesis) allowing to tune polymer properties (like buffering capacity)42. Besides being used as transfection vector, PbAEs has been also applied in others  biomedical areas, such as delivery systems for drugs43;44 or proteins45;46, magnetic  resonance imaging agents47;48, or as scaffold for tissue engineering49;50. Synthesis and main physicochemical properties of poly(b-amino ester)s The poly(b-amino ester)s are easily synthesized by the conjugate addition of a primary  amine or bis(secondary amine) and a diacrylate, in a one-step reaction without  any side product that need be removed through further purification steps. It can be  prepared without solvents, catalysts, or complex protecting group strategies42;51. Depending on the ratio of monomers during the synthesis, poly(b-amino ester)s  can be tailored to have either amine- or diacrylate-terminated chains. An excess of  either diacrylate or amine monomer results in a prevalence of acrylate- or amineterminated  poly(b-amino ester)s, respectively52;53. The synthesis is performed either neat (solvent free) or in anhydrous organic  solvents to mitigate hydrolytic degradation during synthesis42;54. Normally, experiments  using solvents occur at lower temperature and over long periods of time  compared to solvent-free formulations. Table 1.3 summarizes the main reactions for  the synthesis of PbAE and the obtained properties such as molecular weight, polydispersity  index (à ), solvent solubility or yield. The most common solvents used are dimethylsulfoxide (DMSO), chloroform  (CHCl3), or dichloromethane (CH2Cl2)57. However, others solvents have also been  used, such as methanol, N,N-dimethylformamide (DMF) or N,N-dimethylacetamide  (DMA)59;61–63. The solvent used has influence on the final molecular weight of the  PbAE. For example, the use of CH2Cl2 typically yields higher molecular weight  polymer compared to THF42. On the other hand, solvent-free polymerizations maximize monomer concentrations,  thus favoring the intermolecular addition over intramolecular cyclization reaction64. The absence of solvent also allows rising temperature resulting in a higher  reaction rate and a lower viscosity of the reacting mixture, assisting to compensate  the higher viscosity found on the solvent-free systems. The combination between  increased monomer concentration and reaction temperature resulting in a reduction  in the reaction time64. The solvent-free reactions also allows the generation of higher  molecular weight polymers, besides increasing the reaction rate and obviating the  solvent removal step53;64. After polymerization, PbAE can be precipitated, normally in cold diethyl ether,  hexane42, ether65 or ethyl ether58 and/or then dried under vacuum57;65. Frequently, PbAEs are immediately used or stored in the cold conditions (4 _C52;66;67, 0 _C62, or  -20 _C68–70). Some PbAEs should be also kept airproof due to its strong moisture  absorption ability and easy degradation71. Concerning to the biodegradation and biocompatibility, PbAEs have been shown  generally to possess low cytotoxicity and good biocompatibility42;52;61;55;72. Different  studies have suggested that PbAEs are significantly less toxic than currently available  cationic polymers, such as, PEI and PLL51;64. Nevertheless, the increase of the  number of carbons in the backbone or side chain is associated to the increase of the  cytotoxicity73. PbAE degrade under physiological conditions via hydrolysis of their  backbone ester bonds to yield small molecular weight b-amino acids biologically  inert derivatives42;51;55;74. Some results revealed that the degradation rate of poly(b  amino ester)s was highly dependent on the hydrophilicity of the polymer, i.e., the  more hydrophilic the polymer is, the faster the degradation occurs75;76. In Table 1.4 are summarized the main characteristic of PbAEs which make them  a promising polymeric non-viral vector for gene delivery. Combinatorial libraries a fast and efficient way to evaluate different poly(bamino ester)s A fast and efficient way to study the relationships between structure and function  in particular material that could be prepared with different reagents is using combinatorial  libraries. Due to promising preliminar results of PbAEs as non-viral vectors,  Langer research group reported a parallel approach for the synthesis of hundreds of  PbAEs with different structures and the application of these libraries to a rapid and  high throughput identification of new transfection reagents and structure-function trends. For this purpose, major contributions have been reported52;53;57;66;67;72;75;77;78  not only exploring the possible structure/function relationships, but also imposing  an assortment of monomers (amines were denoted by numbers and acrylates by latin  alphabet letters) used in order to facilitate cataloging of different PbAEs (Table 1.5  and Tables A.1 and A.2 (Appendix A)). The first initial library screening was synthesized in 2001 by Lynn51. 140 Different  PbAEs from 7 diacrylates and 20 amines were prepared with molecular weights  between 2,000 and 50,000 g.mol-1. From this, polymers C93 (Mw = 3180 g.mol-1) and  G28 (Mw = 9170 g.mol-1) revealing transfection levels 4-8 times higher than control  experiments employing PEI. At same time, it was observed that for transfection efficiency,  high molecular weight was not an important parameter. This work was then  completed in 2003 by Akinc57, where biophysical properties and the ability of each  polymer/DNA complex to overcome important cellular barriers to gene deliver were investigated. As previous experiments, complexes formed from polymers C93 and  G28, revealed higher levels of internalization compared to †naked† DNA, displaying  18- and 32-fold more internalization, respectively. In contrast, the majority of the  polyplexes were found to be uptake-limited. Regarding d iameter and zeta potential,  out of 10 polymer/DNA complexes with the highest internalization rates, all  had diameters lower than 250 nm and 9 had positive zeta potentials. By measuring  the pH environment of delivered DNA through fluorescence-based flow cytometry  protocol using plasmid DNA covalently labeled with fluorescein (pH sensitive) and  Cy5 (pH insensitive) it was possible to investigate the lysosomal trafficking of the  polyplexes. The results demonstrated that complexes based on polymers C93 and  G28 were found to have near neutral pH measurements, indicating that they were  able to avoid acidic lysosomal trafficking. In the same year, Akinc64 studied the  effect of polymer molecular weight, polymer chain end-group, and polymer/DNA  ratios on in vitro gene delivery. For this purpose, 12 different structures were synthesized  based only in two different PbAE (C28 prepared from 1,4-butanediol diacrylate  and 1-aminobutanol and E28 prepared from 1,6 -hexanediol diacrylate and  1-aminobutanol) (Figure 1.6.) These structures were synthesized by varying amine/diacrylate stoichiometric ratios, resulting in PbAEs with either acrylate or amine end-groups and with molecular  weights ranging from 3,350 to 18,000 g.mol-1. Polymers were then tested, using high  throughput methods, at nine different polymer/DNA ratios between 10/1 (w/w)  and 150/1 (w/w). Concerning terminal groups, it was found that amino-terminated  polymers transfected cells more effectively than acrylate-terminated polymers. In  contrast, none of the acrylate terminated PbAEs mediated appreciable levels of  transfection activity under any of the assessed conditions. These findings suggest that end-chains of PbAE have crucial importance in transfection activity. Concerning  molecular weight effect, highest levels of transfection occurred using the higher  molecular weight samples of both amine-terminated C28 (Mw _13100 g.mol-1 and  E28 (Mw _13400 g.mol-1). Regarding the optimal polymer/DNA ratios for these   polymers, it was observed a markedly difference, 150/1 (w/w) for C28 and 30/1 for  E28. These results highlighted the importance of polymer molecular weight, polymer/DNA ratio, and the chain end-groups in gene transfection activity. Moreover, it  has found the fact that two similar polymer structures, differing only by two carbons  in the repeating unit, have different optimal transfection parameters emphasizing  the usefulness of library screening to perform these optimizations for each unique  polymer structure. Meanwhile, in 2003, Anderson52 described, for the first time,  a high-throughput and semi-automated methodology using fluid-handling systems  for the synthesis and screening of a library of PbAEs to be used as gene carrier. A crucial feature of these methods was that all process of synthesis, storage, and  cell-based assays were performed without removing solvent (DMSO). By using these  methods, it was possible to synthesize a library of 2350 structurally unique, degradable  and cationic polymers in a single day and then test those as transfection reagent  at a rate of _1000 per day. Among PbAEs tested, it was identified 46 polymers  that transfect in COS-7 as good as or better than PEI. The common characteristic  among them was the use of a hydrophobic diacrylate monomer. Moreover, in the  hit structures mono- or dialcohol side groups and linear, bis(secondary amines) are  over represented. From data obtained from this library, Anderson67, in 2004, continued  his study developing a new polymer library of >500 PbAE using monomers  that led higher transfection efficiency in the previous studies and optimizing their  polymerization conditions. The top performing polyplexes were asses sed by using  an in vitro high-throughput transfection efficiency and cytotoxicity assays at different N/P ratios. As previously observed, the most promising polymers are based on  hydrophobic acrylates and amines with alcohol groups. Among those, C32 stood  out due to higher transfection activity with no associated cytotoxicity. The efficiency  to deliver DNA was evaluated in mice after intra-tumoral (i.t.) and intra-muscular  (i.m.) injection. The results revealed important differences. While by i.t injection  C32 delivered DNA 4-fold better than jetPEI R , a commercial polymeric non-viral  vector, by i.m. administration transfection was rarely observed. C32 was then assessed  for DNA construct encoding the DT-A (DT-A DNA) deliver to cells in culture  and to xenografts derived from androgen-sensitive human prostate adenocarcinoma  cells (LNCaP). Results showed that DT-A DNA was successfully delivered and the  protein expressed in tumor cells in culture. In hu man xenografts, the growth was  suppressed in 40% of treated tumors. The fact of C32 is non-toxic and it is able to  transfect efficiently tumors locally and transfects healthy muscle poorly turned it as  a promising carrier for the local treatment of cancer. From here, a panoply of results based in PbAE combinatorial library appeared. In  2005, Anderson53, prepared a new library of 486 second-generation PbAE based on  polymers with 70 different primary structures and with different molecular weights. These 70 polymers were synthesized using monomers previously identified as common  to effective gene delivery polymers. This library was then characterized by  molecular weight of polymers, particle size, surface charge, optimal polymer/DNA  ratio and transfection efficiency in COS-7 of polymer/DNA complexes. Results  showed that from 70 polymers with primary structures, 20 possess transfection activities  as good as or better than Lipofectamine R 2000, one of the most effective commercially  available lipid reagents. Results also revealed that, in general, the most  effective polymers/DNA complexes had In 2006, Green79, synthesized, on a larger scale and at a range of molecular  weights, the top 486 of 2350 PbAEs previously assessed52 and studied their ability to  deliver DNA. These PbAEs were tested, firstly, on the basis of transfection efficacy in  COS-7 cells in serum-free conditions, and then, the 11 of the best-performing PbAEs  structures were further analyzed. The transfection conditions were optimized in human  umbilical vein endothelial cells (HUVECs) in the presence of serum. In this  study, the influence of the factors like polymer structure and molecular weight, and  biophysical properties of the polyplexes (such as, particle size, zeta potential, and  particle stability throughout time) were studied. The results showed that many of  the polyplexes formed have identical biophysical properties in the presence of buffer,  but, when in the presence of serum proteins their biophysical properties changed differentially,  influencing the transfection ac tivity. Concerning to the size, the results  showed that in spite of all vectors condensed DNA into small particles below 150 nm  in buffer, only a few, such as C32, JJ32 and E28, formed small (_200 nm) and stable  particles in serum. C32, JJ32 and E28 revealed also high transfection activity both  in the absence of serum in COS-7 cell line as in the presence of serum in HUVEC  cell line. Moreover, C32 transfected HUVECs in the presence of serum significantly  higher than jetPEI R and Lipofectamine R 2000, the two top commercially available  transfection reagents. The 3 mentioned PbAEs share a nearly identical structure. The acrylate monomers of these polymers, C, JJ, and E, differ by only their carbon  chain lengths (4, 5, and 6 carbons, respectively). Similarly, amines 20, 28, and 32  differ also by only the length of their carbon chain (3, 4, and 5 carbons, respectively). For example, polymers prepared with the same acrylate monomer (C) in which itwas increased the length of the carbons chain of the amine monomer resulted in  an increased transfection efficacy (C32 (5 carbons) > C28 (4 carbons) > C20 (3 carbons))  of these polymers-based polyplexes. Interestingly, this study reinforced C32  as the lead PbAE vector and revealed other potential two, JJ28 and E28, which previously  showedto be poor vectors. On the other hand, C28 and U28, previously  recognized as an efficient transfection reagent, were found to transfect inefficiently  HUVEC in serum. By constructing a new library of end-modified PbAE, the research  was continued78 in order to understand the structure-function relationship  of terminal modification of PbAE in transfection activity. For this purpose, it was  used twelve different amine capping reagents to end-modify C32, D60 and C20. The  choice of these 3 PbAEs was based in their transfection activity: C32, the most effective; D60, an effective transfection reagent with a significantly different structure  from that of C32; and, C20, a poor transfection reagent but with similar structure  to C32 differing only in the length of the amine monomer. The results showed  that some PbAEs-based vectors (C32-103 and C32-117) were able to deliver DNA by  approximately two orders of magnitude higher than unmodified C32, PEI (25,000  g.mol-1) or Lipofectamine R2000, and, at levels comparable to adenovirus at a reasonably  high level of infectivity (multiplicity of infection = 100). Once again, it was  demonstrated that small structural changes influence greatly gene delivery, from biophysical  properties (such as, DNA binding affinity, particle size, intracellular DNA  uptake) until final protein expression. From these 3 polymers assessed, C20 was the  one who transfected cells much less effectively, although it has seen a remarkably  improvement with end-modifications. As expected, C 32-based polyplexes, based on  C32-103 and C32-117, revealed the higher transfection efficiency enhancing cellular  DNA uptake up to five-fold compared to unmodified C32. Interestingly, and in a  general way, terminal modifications of C32 with primary alkyl diamines were more  effective than those with PEG spacers, revealing that a degree of hydrophobicity at  the chain ends is an added value for these polymers. Another interesting fact in terminal  modification of C32 was that at least a three carbon spacer between terminal  amines is necessary to obtain an efficient gene delivery. For example, results showed  that C32-103 transfection efficiency is 130- and 300-fold higher than C32-102 on the  COS-7 and HepG2 cell lines, respectively. As the molecular weight was the same,  this result demonstrated the critical role of the chain ends in transfection activity. In order to better understand the role of the chain ends in transfection efficiency  a new library of end-modified C32 was synthesized by Zugates80 in 2007 using 37  different amine molecules to end-modify the PbAE. In a general way, it was observed  that polymers end-capped with hydrophilic amine end groups containing  hydroxyls or additional amines led to higher transfection efficiency. On the other  hand, terminal-modifications with hydrophobic amines containing alkyl chains or  aromatic rings proved to be much less effective. Concerning to cytotoxicity, terminal  modification with primary monoamine reagents (independently of functional group  extending from the amine, such as aromatic, alkyl, hydroxyl, secondary and tertiary

Hinduism believes that the universe existed first, and then the divine spirit awoke within it and seeded itself and formed Brahma, who is the God. Hinduism, similar to Buddhism, has an evolutionary perspective of life. Both religions accept Darwin’s evolution theory. Over half of the world’s population believes in these religions. They believe the world was created by God in six days, which was recorded in the first chapter of Genesis of the Bible. God created the heaven and earth in the beginning and He created day and night on the first day. Then He separated water from the sky on the second day. On the third day God created vegetation, and on the fourth day He created the Sun, the moon and stars to govern day and night. On the fifth day, God created living creatures that live in the water and fly in the sky. One the sixth day, God made the land to produce living creatures on land, and He said â€Å"let us make man in our image, in our likeness, and let them rule over the fish of the sea and the birds of the air, over the livestock, over all the earth and overall the creatures that move along the ground.† God created man in his own image, and He told man to be fruitful and multiply. On the seventh day, God rested from all the creation he had done. What is evolution? The Merriam-Webster Dictionary has many definition of the term â€Å"evolution† (http://www.m-w.com/). The one definition that this paper is mainly referring to is as follows: â€Å"a theory that the various types of animals and plants have their origin in other preexisting types and that the distinguishable differences are due to modifications in successive generations.† Evolution is tightly associated with Charles Darwin. The British naturalist was under the influence of Charles Lyell, who argued in his book Principles of Geology that the Earth was shaped by accumulative, gradual changes instead of great catastrophes. Darwin traveled with the Beagle around the world and collected specimen. He published his book Origin of species in 1859. The two major themes or Darwin’s theory are â€Å"Descent with Modification† and â€Å"Natural selection and adaptation.
